Los grupos indígenas más numerosos en las islas eran los taínos, los ciboneyes y los caribes. En el año 1517, poco después del comienzo de la conquista, ya era alto el porcentaje de indígenas muertos, debido a conflictos violentos y enfermedades traídas por los europeos. Por lo tanto, la influencia de las culturas indígenas en el Caribe es mínima, en comparación con países como Perú, México y Bolivia.
Debido a la falta de indígenas durante la época colonial, los europeos trajeron a esclavos africanos para trabajar en las islas. Se estima que alrededor de 2 millones de africanos fueron traídos involuntariamente durante la época colonial. La mayoría de estos hombres y mujeres trabajaba en la cosecha de la caña de azúcar, un producto agrícola sumamente importante tanto en Cuba como en la República Dominicana y Puerto Rico. La esclavitud en Cuba no fue abolida hasta 1886, lo que convierte a la isla en el penúltimo país del continente americano en abolirla, siendo Brasil el último.
La presencia africana en las islas caribeñas desarrolló una nueva cultura híbrida, donde se amalgamaron los rasgos culturales típicos de las culturas africanas y las culturas europeas. En Cuba, por ejemplo, la mezcla cultural de las tradiciones africanas con las españolas produjo lo que se conoce como la santería, una religión sincrética que mezcla rasgos de rituales católicos y yorubas.
